Jesse Spencer was born on February 12, 1979 in Melbourne, Australia. The son of Robin Spencer and radiologist Rodney Spencer, he has three siblings, Taney Spencer, Luke Spencer and Polly Spencer.
Spencer attended Canterbury Primary School, Malvern Central School and the private Scots College. While still a student, he auditioned for the long-running soap opera Neighbors.
